Classification

Synthetic pentadecapeptide; large animal literature, minimal human data

Structure and chemistry

BPC-157 is the pentadecapeptide Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val (GEPPPGKPADDAGLV), molecular formula C62H98N16O22, molecular weight approximately 1,419.5 daltons, CAS 137525-51-0. Roughly 27 percent of its residues are proline, which makes the chain unusually rigid and is credited with its stability in water and in gastric juice. It is described as the N-terminal portion of a larger protein reported in mammalian gastric juice. Also known as bepecin, PL 14736 and PL-10.

What the published research shows

The preclinical record is genuinely large — three decades of rodent work across gastrointestinal, tendon, ligament, muscle, bone, vascular and central nervous system injury models, much of it from one Croatian research group. The human record is not. Only a small number of pilot human studies exist, and no controlled clinical trial has validated any indication. A clinical program under the designation PL 14736 reached Phase 2 in inflammatory bowel disease and was not carried through to approval.

Mechanism of action

Reported mechanisms center on nitric oxide signalling through a VEGF-independent route involving Src kinase, and on promotion of angiogenesis linked to the VEGFR2 receptor. An important and unresolved point: no receptor for BPC-157 has been identified, and its sequence has not been located in the human genome. Whether the peptide is genuinely produced by the human body remains scientifically contested rather than settled.

Development history

BPC-157 began as a hypothesis rather than a molecule. Predrag Sikirić, then a medical student at the University of Zagreb, reasoned in 1975 that if the stomach withstands acid and stress it must make its own protective substance. His group spent years collecting gastric juice from hospitals and slaughterhouses and screening it for protective activity, reporting isolation of the active fragment by the end of the 1980s.

Current regulatory status

BPC-157 is not approved by the FDA or any other regulator for any indication anywhere in the world. In September 2023 FDA placed BPC-157 in Category 2 of its interim bulk drug substances list for pharmacy compounding; the nomination was later withdrawn.

Analytical testing

Identity should be confirmed by mass spectrometry against a certified reference standard. Two practical points: the proline-rich sequence makes synthesis prone to deletion sequences, so related peptide impurities are a realistic concern that an area-percentage figure may obscure; and net peptide content differs from chromatographic purity, since acetate counterion and residual water can account for a substantial fraction of vial mass.

Research use only. MontanaPep supplies BPC157 exclusively for in-vitro laboratory research. It is not sold as a drug, food, cosmetic or supplement and is not for human or veterinary use. This page is a reference summary of published literature and regulatory records. Material is produced by pharmaceutical API manufacturers; how that process works is described in our research library.
