Classification
Mitochondrial-derived peptide encoded within the mitochondrial genome
Structure and chemistry
Humanin is a 24-amino-acid peptide whose open reading frame lies within the mitochondrial 16S ribosomal RNA gene. When translated in the cytoplasm from a nuclear-encoded copy it appears as a 21-residue form. It belongs to the small family of mitochondrial-derived peptides that also includes MOTS-c, which is in this catalog.
What the published research shows
Humanin was identified in 2001 by Yuichi Hashimoto and colleagues in surviving neurons from the occipital lobe of an Alzheimer disease patient, in a screen for factors protecting against amyloid-beta toxicity. Subsequent work covers cytoprotection, insulin sensitivity, and the consistent observation that circulating humanin declines with age. The literature is overwhelmingly cell-culture and rodent; no controlled human trials have been published.
Mechanism of action
Reported to act at a trimeric cell-surface receptor complex involving CNTFR, WSX-1 and gp130, and to interfere with pro-apoptotic signalling through BAX. It is also studied as a retrograde signal carrying information from mitochondria to the nucleus.
Development history
An academic research compound with no clinical development programme.
Current regulatory status
Not approved by the FDA. Supplied for laboratory research use only.
Analytical testing
Identity by mass spectrometry, purity by HPLC. Specify whether the 24-residue or 21-residue form is intended, as both circulate in the literature.
