Classification
Synthetic cyclic alpha-MSH analogue
Structure and chemistry
Melanotan II (MT-II) is a cyclic, lactam-bridged heptapeptide analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), with the published sequence Ac-Nle4-Asp5-His6-D-Phe7-Arg8-Trp9-Lys10-alpha-MSH(4-10)-NH2. It is chemically distinct from the earlier linear analogue melanotan-I (also called afamelanotide, or NDP-alpha-MSH), which the same research group developed first.
What the published research shows
Published research spans early receptor-binding and adenylate-cyclase pharmacology, 1994 preformulation and stability chemistry, a 1996 human pilot pharmacology study, and a 1998 double-blind trial in men with psychogenic erectile dysfunction that first documented MT-II's erection-inducing effect.
Mechanism of action
A non-selective melanocortin receptor agonist, activating MC1R (associated with skin pigmentation) along with MC3R and MC4R (associated with appetite and sexual-arousal pathways), which is why its activity extends well beyond pigmentation.
Development history
Developed at the University of Arizona in the late 1980s and early 1990s by M.E. Hadley, V.J. Hruby and colleagues, originally as a candidate chemopreventive sunless-tanning agent intended to reduce UV-induced skin cancer risk.
Current regulatory status
Melanotan II has never been approved by the FDA or any other regulatory authority for any use. Melanotan II was nominated to FDA’s bulk drug substances list for pharmacy compounding; the nomination was withdrawn.
Analytical testing
Identity and purity should be evaluated with methods appropriate to the claimed material, such as HPLC and mass spectrometry. HPLC area percentage alone does not establish molecular identity or net peptide content.
