Classification
Synthetic cyclic melanocortin-receptor agonist
Structure and chemistry
PT-141 (bremelanotide) is a cyclic heptapeptide, Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, structurally derived from Melanotan II and described in the literature as an active MT-II metabolite, chemically modified to favor MC4 receptor activity over the MC1 receptor activity responsible for tanning.
What the published research shows
Published research spans the melanocortin agonist chemistry program at Palatin Technologies, mechanistic and neurobiological studies of MC4R and sexual response, an intranasal-formulation clinical program, and the Phase 3 RECONNECT trials that supported FDA approval as Vyleesi.
Mechanism of action
A melanocortin receptor agonist with activity at MC4R, a receptor implicated in central nervous system pathways governing sexual arousal and desire, distinguishing its mechanism from peripheral vascular drugs such as PDE5 inhibitors.
Development history
Developed by Palatin Technologies after pro-sexual side effects, including spontaneous erections, were observed during early clinical work on Melanotan II for tanning. Early development used an intranasal formulation; the FDA placed a clinical hold on that program over blood-pressure concerns, and Palatin moved to a lower, fixed-dose subcutaneous injection.
Current regulatory status
The bremelanotide molecule is an FDA-approved medicine (Vyleesi), approved in June 2019 specifically for acquired, generalized hypoactive sexual desire disorder (HSDD) in premenopausal women — not for general use or for men. Catalog material sold as PT-141 is a distinct research-grade item and should be verified analytically rather than assumed equivalent to the approved drug product.
Analytical testing
Identity and purity should be evaluated with methods appropriate to the claimed material, such as HPLC and mass spectrometry. HPLC area percentage alone does not establish molecular identity or net peptide content.
