Classification
Endogenous 28-residue neuropeptide of the secretin/glucagon superfamily
Structure and chemistry
Vasoactive intestinal peptide is a 28-amino-acid, C-terminally amidated peptide: His-Ser-Asp-Ala-Val-Phe-Thr-Asp-Asn-Tyr-Thr-Arg-Leu-Arg-Lys-Gln-Met-Ala-Val-Lys-Lys-Tyr-Leu-Asn-Ser-Ile-Leu-Asn-NH2. The C-terminal amide is part of the molecule’s identity, not an optional detail. Human VIP is identical in sequence to bovine, porcine and rat VIP, and the peptide is well conserved across vertebrates — among mammals other than guinea pigs, sequence similarity is at least 85%.
What the published research shows
VIP has a large, well-established research literature covering gastrointestinal physiology, vascular biology, neurotransmission and immune regulation. Preclinical work has shown therapeutic effects in inflammatory models including experimental colitis, sepsis and collagen-induced arthritis, and a clinical trial reported immunoregulatory effects in patients with pulmonary sarcoidosis. The synthetic form, aviptadil, has been investigated in human trials, though VIP is not an FDA-approved standalone medicine.
Mechanism of action
VIP acts through the VPAC1 and VPAC2 G-protein-coupled receptors, stimulating adenylate cyclase and raising intracellular cAMP. It is a potent vasodilator and smooth muscle relaxant acting on peripheral, pulmonary, coronary and cerebral vascular beds, stimulates pancreatic and biliary secretion, and has documented immunomodulatory effects including on lymphocyte proliferation and migration. It crosses the blood-brain barrier largely by transmembrane diffusion.
Development history
VIP was not designed. Sami I. Said and Viktor Mutt isolated it from porcine intestine in 1970 while tracking a vasodilator activity through successive purification steps — the activity itself is what guided the isolation, and is what gave the peptide its name.
Current regulatory status
VIP itself is an endogenous human hormone. Its synthetic form, aviptadil, has been studied in clinical trials, but VIP is not an FDA-approved standalone medicine in the United States. It has not been placed on FDA’s bulk drug substances lists. Catalog material sold as VIP is a research-grade item.
Analytical testing
Two features matter for identity here. The C-terminal amidation must be confirmed — a free-acid version is a different molecule with altered activity, and it differs by only about 1 dalton, so the mass-spectrometry method must be capable of resolving that. VIP also contains a methionine residue, which is oxidation-prone, making oxidized variants a realistic impurity class that a purity percentage alone does not characterize.
