Classification
Modified GHRH(1-29) analogue, short-acting; also called Mod GRF 1-29
Structure and chemistry
CJC-1295 without DAC is a 29-residue analogue of the first 29 amino acids of growth-hormone-releasing hormone, GHRH(1-29), carrying four substitutions: D-Ala at position 2, Gln at 8, Ala at 15, and Leu at 27. These changes resist degradation by dipeptidyl peptidase-IV. It is also known as Mod GRF 1-29 or CJC-1295 DAC-free. It is chemically distinct both from DAC-modified CJC-1295 (which carries an additional albumin-binding lysine linker) and from sermorelin (which is unmodified GHRH(1-29)).
What the published research shows
Published human clinical research on CJC-1295 was conducted using the DAC-modified form. The no-DAC form does not have its own dedicated published human clinical trial program, and results from DAC-form trials cannot be transferred to it: the two have half-lives differing by roughly two orders of magnitude and therefore entirely different pharmacokinetic behaviour.
Mechanism of action
A GHRH receptor agonist that stimulates growth hormone release from the anterior pituitary. Without the DAC albumin-binding modification, its plasma half-life is roughly 30 minutes, producing a short pulse of GH release rather than sustained elevation.
Development history
Derives from the same ConjuChem Biotechnologies GHRH program that produced DAC-modified CJC-1295. The tetrasubstituted GRF(1-29) backbone is the shared component; the no-DAC form is that backbone without the drug affinity complex.
Current regulatory status
Not approved by the FDA or any regulator for any indication. CJC-1295 was nominated to FDA’s bulk drug substances list for pharmacy compounding; the nomination was withdrawn.
Analytical testing
The COA should clearly identify the no-DAC form and use a mass-spectrometry-based identity method able to distinguish it from both the DAC-modified form and unmodified sermorelin, since all three share a similar backbone but differ in mass.
