Classification
Truncated IGF-1 analogue lacking the first three N-terminal residues
Structure and chemistry
IGF-DES is des-(1-3)-IGF-1: native human IGF-1 with the first three amino acids of the N-terminus — glycine, proline and glutamic acid — removed, leaving a 67-residue protein. The critical residue lost is the glutamic acid at position 3, the same residue that IGF-1 LR3 replaces with arginine. The two analogues therefore solve the same problem by opposite means: LR3 substitutes that glutamate, DES deletes it.
What the published research shows
The foundational work is a careful truncation series that remains unusually informative. Researchers synthesized IGF-1 analogues shortened by one, two, three, four and five residues and measured protein-synthesis potency in rat L6 myoblasts, which produces a clean picture of exactly which residues matter and why.
Mechanism of action
Removing the N-terminal tripeptide, and specifically the glutamate at position 3, largely abolishes IGF binding protein recognition while leaving type 1 IGF receptor binding intact. The result is a molecule whose activity is not attenuated by binding proteins secreted in the local environment. The truncation series shows this is a knife edge: cutting one residue too far collapses activity entirely.
Development history
Developed within the same Australian IGF-1 analogue research program that produced Long [Arg3] IGF-1, as part of systematically mapping the functional role of the IGF-1 N-terminus.
Current regulatory status
IGF-DES is not approved by any regulator for any indication and has no human clinical trial record. It has not been placed on FDA’s bulk drug substances lists.
Analytical testing
Identity requires confirming that exactly three residues have been removed. Mass spectrometry distinguishes des-(1-3) from des-(1-2) and des-(1-4) forms, and that distinction matters enormously: the published data show a difference of several hundredfold in potency across neighbouring truncation states, so a mis-truncated product is not a minor impurity but a functionally different molecule.
