Classification
Synthetic hexapeptide growth hormone secretagogue; the founding compound of its class
Structure and chemistry
GHRP-6 is the hexapeptide His-D-Trp-Ala-Trp-D-Phe-Lys-NH2, molecular weight approximately 873 daltons. It contains two D-configuration residues and a C-terminal amide. Despite being derived structurally from met-enkephalin analogues, it has no opioid activity.
What the published research shows
GHRP-6 has a broad preclinical literature across multiple species — the original work demonstrated dose-related GH release in chickens, rats, monkeys, lambs and calves, as well as in humans. Human research has largely been physiological and pharmacological rather than therapeutic; GHRP-6 was never developed to approval for any indication.
Mechanism of action
An agonist at the growth hormone secretagogue receptor GHS-R1a, the receptor whose natural ligand ghrelin was not identified until 1999. GHRP-6 acts synergistically with GHRH in vivo. Among the GHRPs it produces the strongest appetite stimulation, and at higher doses it also modestly raises cortisol and prolactin.
Development history
Discovered by the American endocrinologist Cyril Y. Bowers at Tulane University School of Medicine, New Orleans, working with the computational chemist Frank Momany. The origin was serendipitous: Bowers was modifying enkephalins, the body’s natural opioid peptides, when he found that certain analogues triggered growth hormone release from pituitary cells while having no analgesic activity at all.
Current regulatory status
Not approved by the FDA or any other regulator for any indication. FDA placed GHRP-6 in Category 2 of the 503B interim bulk drug substances list in September 2023.
Analytical testing
Identity should be confirmed by mass spectrometry against a certified reference standard, with attention to the D-Trp and D-Phe stereochemistry and the C-terminal amide, none of which an HPLC purity figure establishes.
