Classification
C-terminal fragment of human growth hormone; a discontinued clinical candidate
Structure and chemistry
A 16-residue peptide corresponding to amino acids 176 to 191 at the carboxyl terminus of the 191-residue human growth hormone molecule. Critically, it lacks the regions of the parent hormone responsible for receptor-mediated growth signalling, which is the entire premise of the compound.
What the published research shows
The core literature is rodent work from the 1990s and 2000s, much of it from Frank Ng and colleagues at Monash University. Reported findings include reduced body-weight gain in obese Zucker rats and increased lipolytic activity in adipose tissue, without the rise in insulin-like growth factor 1 that accompanies full-length growth hormone. Human work is limited and the clinical programme did not succeed.
Mechanism of action
Proposed to act through beta-3 adrenergic receptor signalling in adipocytes rather than through the growth hormone receptor. This is the basis of the claim that it separates metabolic effects from growth effects — a separation demonstrated in rodents but never established in humans.
Development history
Developed by Metabolic Pharmaceuticals in Australia as an obesity candidate under the designation AOD9604-adjacent chemistry. The related fragment AOD 9604 advanced further, failed to beat placebo in a Phase 2b trial, and the programme was discontinued. No successor reached approval.
Current regulatory status
Not approved by the FDA or any other regulator for any indication. Supplied here for laboratory research use only.
Analytical testing
Identity by mass spectrometry against the expected molecular weight, purity by HPLC. Because the sequence is a fragment of a much larger well-known protein, confirming the correct fragment rather than a related one matters.
