Classification

Splice variant of insulin-like growth factor 1; the mechano-responsive isoform

Structure and chemistry

Mechano-growth factor is an alternatively spliced isoform of IGF-1, also designated IGF-1Ec in humans. A reading-frame shift arising from a 52-base-pair insertion in exon 5 produces a distinct C-terminal E-domain. Research material sold under this name is typically the 24-residue E-domain peptide rather than the complete protein, which is an important distinction when interpreting literature.

What the published research shows

MGF was characterised in skeletal muscle subjected to mechanical loading and damage, where expression rises transiently before the systemic IGF-1Ea isoform takes over. Preclinical work has examined satellite cell activation and muscle repair. Research has since extended into growth-plate cartilage and, in aged mice, neurogenesis. A recurring methodological problem is that the unmodified E-domain peptide has a very short serum half-life, which complicates in-vivo interpretation and is the reason pegylated versions exist.

Mechanism of action

The E-domain peptide is reported to act on muscle satellite cells through a pathway distinct from the IGF-1 receptor. The receptor responsible has not been definitively identified, which remains a significant gap in the mechanistic account.

Development history

An academic research compound arising from work by Geoffrey Goldspink and colleagues. No clinical development programme has reached approval.

Current regulatory status

Not approved by the FDA for any indication. Supplied for laboratory research use only.

Analytical testing

HPLC purity and mass spectrometry identity. Specify whether material is the E-domain peptide or pegylated MGF, since the two are not interchangeable in the literature.

Research use only. MontanaPep supplies MGF exclusively for in-vitro laboratory research. It is not sold as a drug, food, cosmetic or supplement and is not for human or veterinary use. This page is a reference summary of published literature and regulatory records. Material is produced by pharmaceutical API manufacturers; how that process works is described in our research library.
