Classification

Synthetic seven-residue fragment of spadin; a TREK-1 channel blocker

Structure and chemistry

PE-22-28 corresponds to residues 22 to 28 of spadin, itself derived from the propeptide released during maturation of sortilin. The shortened sequence was designed to retain affinity for the target while improving stability over the parent 12-residue molecule.

What the published research shows

The work centres on the TREK-1 two-pore-domain potassium channel as an antidepressant target, following the finding that TREK-1 knockout mice display an antidepressant-like phenotype. Rodent studies from Marc Borsotto, Catherine Heurteaux and colleagues at the Institut de Pharmacologie Moléculaire et Cellulaire in France report rapid-onset antidepressant-like behaviour and increases in hippocampal neurogenesis. There is no published human clinical data of any kind.

Mechanism of action

Blocks the TREK-1 channel. This is mechanistically distinct from monoamine reuptake inhibition, and the interest in it stems from the speed of onset reported in animals compared with conventional antidepressants.

Development history

An academic research compound. It has not entered clinical trials and has no registry presence.

Current regulatory status

Not approved by the FDA. Entirely preclinical. Supplied for laboratory research use only.

Analytical testing

Identity by mass spectrometry, purity by HPLC. A short sequence, so mass confirmation is straightforward and should be expected on the COA.

Research use only. MontanaPep supplies PE-22-28 exclusively for in-vitro laboratory research. It is not sold as a drug, food, cosmetic or supplement and is not for human or veterinary use. This page is a reference summary of published literature and regulatory records. Material is produced by pharmaceutical API manufacturers; how that process works is described in our research library.
