Classification
Synthetic tuftsin analogue, registered as a medicine in Russia only
Structure and chemistry
Selank is a heptapeptide with the sequence Thr-Lys-Pro-Arg-Pro-Gly-Pro. Its first four residues reproduce tuftsin, an endogenous immunomodulatory tetrapeptide released by enzymatic cleavage of the immunoglobulin G heavy chain; the C-terminal Pro-Gly-Pro tripeptide is a stabilizing addition that resists enzymatic degradation. This is the same Pro-Gly-Pro stabilization strategy used in Semax, which came from the same institute.
What the published research shows
Published research includes rodent studies of anxiety-related behavior, BDNF expression and GABAergic gene expression, cell-based receptor and transcriptome work, and Russian human clinical studies in generalized anxiety disorder. The principal published human trial used an active benzodiazepine comparator rather than a placebo, and no Western randomized controlled trial has replicated these findings.
Mechanism of action
Reported mechanisms include inhibition of enkephalin degradation (extending the half-life of endogenous enkephalins), modulation of BDNF expression in the hippocampus and prefrontal cortex, effects on the expression of genes involved in GABAergic neurotransmission, and modest modulation of serotonin and dopamine turnover. As a tuftsin analogue it also retains immunomodulatory activity.
Development history
Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences in Moscow, in cooperation with the Zakusov Scientific Research Institute of Pharmacology — the same institutional lineage that produced Semax.
Current regulatory status
Selank is registered as a prescription anxiolytic in Russia only. It has never been approved by the FDA, the EMA, or any regulator outside Russia. Catalog material sold as Selank is a research-grade item and should not be treated as equivalent to the Russian registered drug product.
Analytical testing
HPLC and mass spectrometry provide complementary chromatographic and identity evidence; the exact heptapeptide sequence should be confirmed against a certified reference standard rather than inferred from an HPLC purity figure alone.
